Design Notes

One of the main challenges was designing parts that respond to high pH conditions. Since the overexpressed genes were identified under pH 11 conditions, it was crucial to ensure that any regulatory elements (promoters, enhancers) linked to these genes would remain active and responsive in similar alkaline environments. We needed to evaluate whether the native promoters driving these genes' expression were strong enough under high pH or if additional tuning would be required.
